Sammanfattning

This article reviews the recent landmark genetic studies of common and rare variation shaping the highly polygenic architecture of schizophrenia. Functional alleles have now been identified explaining specific molecular mechanisms implicating excessive complement activity in the development of schizophrenia. Furthermore, we conclude that results from current genome-wide association studies may hold potential therapeutic leads. The impact of multiple environmental exposures is discussed, and the complexity of schizophrenia aetiology is underlined by environmental factors acting alone or in gene-environment interactions.
