P16INK4a og Ki-67 - et alternativ til rescreening av livmorhalspröver med normal cytologi og
positiv HPV-test?

Background: Double immunostaining with CINtec® PLUS can be used to detect the proteins
p16INK4a and Ki-67 in cytological cervix preparations. Detection of both proteins in the same cell is
an indication of deregulated cell cycle and possible precancerous lesions. The purpose of the study
was to determine if double immunostaining of p16INK4a and Ki-67 could be used for localization of
cells that could represent precancerous lesions of the cervix in women with normal Pap smear and
positive HPV DNA test.
Material and methods: 25 cervical specimens, assessed as cytological normal and tested HPV DNA
positive, were used. The method was double immunostaining of p16INK4a and Ki-67 with use of
CINtec® PLUS.
Results: 10 of 25 samples were positive by the double immunostaining, 14 were negative and one
was considered uncertain. 4 of the 10 positive samples contained abnormal cells by cytological
evaluation. These were diagnosed ASC-US and LSIL.
Conclusion: In 4 of 25 samples there were found low-grade cervical dysplasia. Double
immunostaining of p16INK4aand Ki-67 can be used to find cells with abnormalities in samples
assessed as cytological normal and tested HPV DNA positive.
